The championships also acted as the opening round of the Renshaw's Pedal Project Bathurst Cycle Club Road Series.
Ryan completed the 15 kilometre course in 20 minutes and 38 seconds for an average speed of 43.6 kilometres an hour.
Runner-up Craig Hutton also snuck under the 21 minute mark (20:59) while third place in A grade went to Mark Windsor (21:23).
The winning time puts Ryan just 17 seconds shy of the official club record of 20:21 set by Dean Windsor in 2006, although faster unofficial times have been recorded.
"I think it was a reasonable time on the day. Being only 15kms it's pretty much as hard and as fast as you could. I wanted to go out hard and just see if I could hold on," Ryan said.
"I think that's probably the fastest time I've done on that course. I was having a look through the times that have been done previously and from what I could see only one has ever managed to go under 20 and I didn't know that was the case for today. Maybe that could be a goal for the future."
Ryan was quick to talk down his chances of adding more wins to his name across the remainder of the road series this season.
"The time trial was definitely my biggest possibility of getting a win in this series," he said.
"The road races? There's not going to be as much of a possibility for me to be there. I'll do the ones I can but the young guys are too fast for me."
Only a group of around 20 riders have managed to get themselves below the 21 minute mark over the two decades of racing on the Perthville circuit.
B grade was won by Steve Bennett with the third fastest overall time of the day (21:12). Damien Bennett (23:44) was the C grade winner while D grade went the way of Cathy Adams (27:54).
