Riley is completing the run to raise funds for the Chris O'Brien Lifehouse project, a not-for-profit, comprehensive cancer hospital in Camperdown, Sydney.
Riley's run will be leaving the Gooloogong Post Office at 7am and finishing at the Cowra Bowling Club.
"I lost a grandfather last year on the second of May to a brain tumour," Riley said.
Anyone who wants to join him for the full or part of the journey is most welcome but will do so at their own risk.
Two support vehicles will be accompanying the runners, who include staff from Coles in Cowra.
"It's not going to be a cracking hot pace, I'm not going out there to run a fast as you can pace," he said.
If you're not up to covering the full distance Riley said: "People can join the last little bit, from near the Cowra Gun Club, about 5km out of town, if they wish".
The runners should reach this point between 10am and 10.30am on Sunday.
